Mechanism of action: Hygromycin B is an aminoglycoside antibiotic produced by the metabolism of Streptomyces hygroscopicus, which kills bacteria, fungi and higher eukaryotic cells by inhibiting protein synthesis. Hygromycin B inhibits protein synthesis by interfering with 70S ribosome translocation and inducing misreading of mRNA templates. It is currently used to screen and maintain cultured prokaryotic or eukaryotic cells containing a hygromycin resistance gene.
Resistance gene:
Resistance to hygromycin B is derived from a gene encoding hygromycin B phosphotransferase (Hph). Two sources have been discovered so far:
♦ Hph resistance gene produced by Streptomyces hygroscopicus;
♦ Escherichia coli, the Hph resistance gene carried by the plasmid in Klebsiella pneumoniae (commonly used);
Biological applications:
1) screening and maintaining cultured prokaryotic or eukaryotic cells (plant cells and mammalian cells) stably transfected with Hph+ vector;
2) Due to the difference in mode of action, it is often used in combination with G418 (GeneticinTM), ZeocinTM and blasticidin S to screen cell lines stably transfected with two different vectors;

Application concentration:
The working concentration of hygromycin B used to screen stable strains varies depending on cell type, medium, growth conditions, and cellular metabolic rate. The recommended concentration is 50-1000 μg/mL, and the optimal concentration is determined by the kill curve.

Product use:
Use one: kill curve to determine the best kill concentration (for reference only, depending on the personal detection system)
(1) Add untransfected cells to a tissue culture grade 96-well plate at a cell density of about 50-200 cells/well; after the cells are attached, add different concentrations to each well (eg, 50-1000 μg/mL, at least 5 200 μL fresh medium of hygromycin B;
(2) Incubate the cells for 10-14 days at 37 ° C in a 5% CO 2 incubator;
(3) After 5-7 days of culture, replace the new culture medium, and the culture solution contains the corresponding concentration of hygromycin B;
(4) After 10-14 days, cell viability is evaluated using a cell proliferation method such as MTT, CCK-8, etc. The toxic effect can also be determined by measuring the number of cell clones or the percentage confluence. The minimum concentration that is capable of killing all cells in 10-14 days is generally selected as the optimal screening concentration.
Use two: screening stable transfected cells
(1) For adherent cells, directly absorb the transfected cell culture medium in a 60 mm culture dish, then add 5-6 ml of fresh medium containing hygromycin B; for suspension cells, sterile conditions 250 x g, centrifuge for 10 min to remove The old transfected cell culture medium is then suspended in about 5 ml of fresh medium containing hygromycin B;
(2) After 5-7 days, replace the fresh medium containing hygromycin B as above;
(3) re-incubate the cells for 5-7 days;
(4) After 10-14 days of incubation, the culture system contained only viable cells capable of expressing the hygromycin B resistance phenotype. Therefore, after the screening, as in step 1, the fresh medium containing no hygromycin B was replaced.23
