Introduction
The HiPure Fruit RNA Mini Kit provides fast purification of high-quality RNA from Fruits, cell, tissues, and yeast using silica-membrane spin columns with a binding capacity of 100ug RNA. There is no need for phenol/chloroform extractions and time-consuming steps such as CsCl gradient ultracentrifugation, or precipitation with isopropanol or LiCl are eliminated. RNA purified using the HiPure Total RNA Purification System is ready for applications such as RT-PCR, Northern blotting, poly A+ RNA (mRNA) purification, nuclease protection, and in vitro translation.
Procedure
This kit uses silica membrane purification technology, only need to perform simple binding-washing-elution steps.
The sample is lysed and homogenized in the lysis buffer containing guanidine salt. The lysate is transferred to the DNA filter column for centrifugation. The DNA is removed by high-efficiency filtration and the RNA-containing filtrate is collected. Ethanol is added to provide suitable binding conditions. Transfer to the spin column after neutralization and centrifugation.RNA can be selectively bound to the membrane. The contaminants are washed away after three washings. The purified RNA is eluted by RNase Free Water finally.
For some RNA applications that are very sensitive to a small amount of DNA, the on-column DNase digestion process can be carried out during RNA extraction.T degraded DNA fragments and DNase can be removed with the washing solution to obtain high-purity RNA without DNA and DNase contamination.
