This kit is impregnated with dried monoclonal antibody Anti-1.1 and Anti-1.2 for determining blood type.
The antibody in this kit was obtained by injecting specific hybridoma into Balb/C mice.
Anti 1.1 and Anti 1.2 showed the molecular weights of approximately 200 kDa and 80 kDa, respectively, as measured by western blot analysis(Lee et al., plos one,2017).
* Clinical importance of blood types
When canine is transfused with compatible blood type at the first transfusion, the half-life of the blood within the body
is long as 21 days. But, when canine is transfused with incompatible blood type, the half-life of the blood within body
is short, approximately 12 hours. (Bull RW: Antigens, grafts rejections, and transfusions. J An Vet Med Assoc 181:1115,
1982) Thus, the blood typing is crucial for successful transfusion for the first time.
There are 13 blood types for dogs. Especially, type 1.1(DEA1.1), and type 1.2(DEA1.2) are very important for transfusion
*1 : If a negative dog is exposed to 1.1 positive erythrocytes, a strong hemolysin can result acute hemolytic reaction.
On second exposure, immune-mediated hemolytic transfusion reaction results causing removal of transfused
cell in less than 12 hours.
*2 : A previously sensitized negative type dog undergoes permanent red blood cell removed and loss 12 to 24
hours after administration of 1.2 positive erythrocytes.
※ Therefore, transfusion must be carried out according to blood type.
* Test procedure
1. Collect 500ul of canine blood. (Use EDTA tube, or a tube with anti-coagulant)
2. Put one drop of blood using approximately 20-30ul into each well of blood card.
3. Put one drop of PBS(20ul) each well.
4. Mix with stick (15 seconds) (Use different stick in each well)
